Elemento transponível ou transposon, isolado de Bacillus thuringiensis (B.t) e designado como transposon n5401. A invenção também inclui um método de uso deste transposon em um sistema de recombinação específica de sítio para construção de cepas de B.t. recombinantes que contêm gens de proteína de toxina de B.t. inseticida e que são livres de DNA não nativo do B.t.
