Abstract
"SERINA RACEMASE DE MAMÍFEROS". Altos níveis de D-serina ocorrem no cérebro de mamíferos, onde ela parece ser um ligante endógeno do "sítio de glicina" dos receptores de NMDA. Nesta invenção, foi purificada, a partir do cérebro de ratos, uma enzima solúvel que catalisa a racemização direta de L-serina para D-serina. A serina racemase purificada tem um peso molecular de 37 kDa e requer 5'-fosfato de piridoxal para sua atividade. A enzima é altamente seletiva na direção de L-serina, deixando de racemizar qualquer outro aminoácido testado. Identificou-se também seqüências de polinucleotídeos que codificam uma serina racemase de mamíferos, inclusive humana. Os compostos que modulam a atividade da serina racemase de mamíferos são úteis para tratar condições e doenças que envolvem a superestimulação de receptores de NMDA, tais como acidente vascular cerebral e doenças neurodegenerativas.
