Abstract
AbstractThe present invention relates to methods for developing engineered T-cells for immunotherapy that are non-alloreactive. The present invention relates to methods for modifying T-cells by inactivating both genes encoding T-cell receptor and an immune checkpoint gene to unleash the potential of the immune response. This method involves the use of specific rare cutting endonucleases, in particular TALE-nucleases (TAL effector endonuclease) and polynucleotides encoding such polypeptides, to precisely target a selection of key genes in T-cells, which are available from donors or from culture of primary cells. The invention opens the way to standard and affordable adoptive immunotherapy strategies for treating cancer and viral infections. traduçaõ do resumoRESUMOPatente de Invenção: "MÉTODOS PARA MODIFICAR GENETICAMENTE CÉLULAS T ALOGENEICAS E ALTAMENTE ATIVAS PARA IMUNOTERAPIA". A presente invenção refere-se a métodos para desenvolver células T geneticamente modificadas para imunoterapia que são não-alorreativas. A presente invenção refere-se a métodos para modificar células T inativando ambos os genes codificando o receptor de células T e um gene de barreira imunológica para desatrelar o potencial da resposta imunológica. Este método envolve o uso de endonucleases de corte raro específicas, em particular TALE-nucleases (endonuclease efetora de TAL) e polinucleotídeos codificando tais polipeptídios, para atingir com precisão uma seleção de genes essenciais nas células T, que estão disponíveis a partir de doadores e a partir de cultura de células primárias. A invenção abre caminho para estratégias de imunoterapia adotiva tradicionais e acessíveis para o tratamento de câncer e infecções virais.1/2
