Abstract

USOS DE UMA CÃLULA DE SOJA E MÃTODOS DE PIRAMIDAÃÃO DE MÃLTIPLOS CASSETES DE EXPRESSÃO DE INTERESSEA presente invenÃ§Ã£o descreve um mÃ©todo de direcionamento que permite a substituiÃ§Ã£o precisa de cassetes em um locus gÃªnico previamente caracterizado. Uma construÃ§Ã£o de DNA alvo que contÃ©m um par de sÃtios FRT incompatÃveis flanqueando um cassete alvo foi introduzida em soja por meio de transformaÃ§Ã£o biolÃstica regular. Eventos transgÃªnicos que contÃªm uma Ãºnica cÃ³pia completa do sÃtio alvo foram selecionados em seguida retransformados com uma construÃ§Ã£o de DNA doador que contÃ©m o par idÃªntico de sÃtios FRT incompatÃveis flanqueando um cassete doador. A troca precisa de cassetes de DNA ocorreu entre o cassete alvo e o cassete doador por meio de troca de cassetes mediada por recombinase (RMCE), de tal forma que o cassetes doador fosse introduzido no sÃtio genÃ´mico exato ocupado anteriormente pelo cassete alvo. Por meio de RMCE repetida utilizando sÃtios FRT incompatÃveis adicionais, mÃºltiplos grupos de transgenes podem ser piramidados no mesmo locus genÃ´mico.
