Abstract

ANTICORPO BIESPECÍFICO ANTI-TSPAN8/ANTI-CD3 E ANTICORPO ANTI-TSPAN8. Um objetivo da presente invenção é fornecer um anticorpo biespecífico anti-TSPAN8/anti-CD3 e um anticorpo anti-TSPAN8 utilizáveis no tratamento ou prevenção em humanos. Um camundongo produtor de anticorpo monoclonal humano foi imunizado com uma célula cancerígena disseminada peritoneal isolada de um paciente, para obter um anticorpo 16B11 e um anticorpo 16B12 que se ligam seletivamente a uma célula cancerígena disseminada peritoneal. Esses anticorpos eram anticorpos anti-TSPAN8 que se ligam à região das posições de aminoácidos 126 a 155 de TSPAN8 e exibiram forte atividade de ligação a TSPAN8 expresso na célula cancerígena disseminada peritoneal. Além disso, um anticorpo biespecífico anti-TSPAN8(16B11)-anti-CD3 produzido com base na sequência de 16B11 exibiu uma atividade citotóxica contra a célula cancerígena que expressa TSPAN8 in vitro, exerceu uma ação antitumoral em camundongos portadores de células cancerígenas que expressam TSPAN8 in vivo, e prolongaram a vida útil de camundongos modelo de disseminação peritoneal.
