Abstract

TRATAMENTO DE CÂNCER COM DEFICIÊNCIA DE RH. Esta invenção se refere à verificação de que células deficientes em recombinação homóloga (RH) são sensibilizadas à inibição de PARP por (i) inibição catalítica ou ablação genética de 2'-desoxinucleosídeo 5'-fosfato N-hidrolase 1 (DNPH1) ou (ii) administração de um substrato de DNPH1, tal como 5-hidroximetil-desoxiuridina (hmdU). A invenção também se refere à descoberta de que a inibição catalítica ou ablação genética de DNPH1 combinada com a administração de hmdU causa letalidade sintética em células deficientes em RH na ausência de inibição de PARP. São providos métodos e compostos para uso no tratamento de câncer com deficiência de RH.
