Abstract
A invenção fornece métodos de triagem baseada em célula eucariótica para identificar um aptâmero que se liga especificamente a um ligante, ou um ligante que se liga especificamente a um aptâmero, com o uso de um cassete de polinucleotídeo para a regulação da expressão de um gene repórter em que o cassete de polinucleotídeo contém um riboswitch no contexto de um íntron 3' de éxon alternativo de íntron 5'. O riboswitch compreende uma região efetora e um aptâmero de modo que, quando o aptâmero se ligar a um ligante, a expressão de gene repórter ocorra.
