A invenção compreende um novo e inventivo método de conjugação da enzima L-asparaginase com moléculas de polietilenoglicol, preferencialmente polietilenoglicol maleimida (PEG-Mal), através de resíduos de cisteína presentes na superfície da molécula. Adicionalmente, a invenção se refere ao produto obtido a partir deste método e à utilização da enzima conjugada com PEG no tratamento de diferentes tipos de neoplasia, tais como Leucemia Linfoide Aguda.
